What is Granulation and common tablet tooling technology

Granulation is the process of forming granules from powders in pharmaceutical tablet manufacturing. The granulation method used can have a significant impact on the properties and performance of the final tablets. Here are some common types of granulation methods used in tablet manufacturing:

  1.  Wet granulation: This method involves the addition of a liquid binder to the powder mixture to form granules. The mixture is then dried and screened to obtain the desired particle size distribution. Wet granulation is a commonly used method because it produces uniform granules with good flow properties.

  2.  Dry granulation: Also known as slugging, this method involves compressing the powder mixture into large tablets or slugs. The slugs are then broken down into granules using a milling machine. Dry granulation is useful for heat-sensitive materials and can avoid the need for a liquid binder.

  3.  Roller compaction: This dry granulation method involves feeding the powder mixture between two counter-rotating rollers. The rollers compress and density the powder, forming a solid sheet or ribbon. The sheet is then milled to produce granules. Roller compaction is a continuous process that can produce high-quality granules with good flow properties.

  4.  Spray drying: In this method, the powder mixture is dissolved in a solvent and sprayed into a hot air stream. The solvent evaporates, leaving behind dry granules. Spray drying is useful for heat-sensitive materials and can produce very fine and uniform granules.

  5.  Extrusion/spheroidization: This method involves extruding the powder mixture through a die to form cylindrical shapes. The cylinders are then spheroidized in a rotating drum to produce spherical granules. Extrusion/spheroidization can produce uniform, spherical granules with good flow properties.

  The choice of granulation method will depend on the properties of the materials being used, the desired properties of the final tablets, and the equipment available for manufacturing.

 

 Tablet compression is the process of compacting a granulated or powdered drug substance into a tablet form using a tablet press machine. The tablet press compresses the granulated or powdered material into a specific size, shape, and weight. The compressed tablets can then be coated or packaged for use.

 The tablet compression process involves several steps:

  1.  Pre-compression: The granulated or powdered material is fed into the tablet press and pre-compressed to form a solid mass or slug.

  2.  Main compression: The pre-compressed material is then compressed again at a higher pressure to form the final tablet.

  3.  Ejection: The tablet is ejected from the press and collected in a container.

 The tablet press machine is equipped with several components, including the hopper, feeder, die, punches, and cam tracks. The hopper  used to hold the granulated or powdered material, and the feeder used to control the flow of material into the die cavity. The die cavity determines the size and shape of the tablet, and the punches  used to compress the material into the die cavity.

  Tablet compression is an important step in tablet manufacturing and requires careful control of various parameters, including the compression force, tablet weight, and tablet hardness. The final tablet properties, such as disintegration time, dissolution rate, and stability, can be affected by the compression process.
